Correction by peptide pharmaceuticals of reproductive disorders in female rats caused by prolonged morphine administration

Brief summary

The study purpose was to assess the effect of delta sleep inducing peptide (DSIP) and an ana-logue of the 4-10 fragment of adrenocorticotropic hormone (FAKTH) on reproductive dys-function of female rats caused by a prolonged previous morphine intake. It was found that the administration of morphine to non-inbred female rats for 90 days at a dai-ly dose of 60 mg/kg had a significant effect on the reproductive function of the rats: - a decrease in fecundity of rat females, as evidenced by a decrease in fertility and pregnancy indices, as well as an increase in pre- and post-implantation death indices; - abnormalities in the development of the internal organs of the fetus (expansion of the ventri-cles of the brain, hydronephrosis and hemorrhages in the brain); - a decrease in morphometric indices of embryos bodies, as well as a decrease in the length of the primordia of the bones and the number of ossification centers of the small bones of the dis-tal parts of the front and rear extremities. In turn, the use of peptide pharmaceuticals in selected doses leveled disturbances in the repro-ductive function of female rats, caused by prolonged administration of morphine, as evidenced by: - restoration of fertility, manifested by the normalization of fertility and pregnancy indices, as well as pre- and post-implantation death to the indicators of the control group; - reduction to a minimum of the occurrence of abnormalities in the development of internal organs (expansion of the ventricles of the brain, hydronephrosis and hemorrhages in the brain) in rat embryos; - decrease in the severity of dolichomorphia in rat embryos. It was also established that the use of a fragment of ACTH, in contrast to DSIP, contributed to the restoration of ossification of the small bones of the distal parts of the fore and hind limbs of embryos in rat females that received morphine for a long time before pregnancy.
