Synthetic genes for molecular biology by the example of human placental ribonuclease inhibitor's gene

Brief summary
In this paper we suggest a new approach for chemical synthesis of genes. The human placental ribonuclease inhibitor's gene was obtained by this method. The nucleotide sequence optimisation for expression in Escherichia coli allows to achieve about 4-fold higher yield of protein than in case of native gene. New chromatographic purification strategy without using of immobilized ribonuclease was developed for the protein purification. Gene optimization allows to improve expression both in E.coli and in eucaryotic systems.
