Method for measuring benzo[a]pyrene metabolite concentrations in human urine by gc/ms for the purpose of exposure biomonitoring

Brief summary

The authors developed and tested method for measuring (MM) the metabolite of benzo[a]pyrene (BaP) -3-hydroxybenzo[a]pyrene (3-OH-BaP) in urine. Since benzo[a]pyrene is one of the most carcinogenic polycyclic aromatic hydrocarbons (PAHs), the measurement of its metabolites will provide the most accurate assessment of PAH exposure by employees of enterprises, including those working in especially hazardous working conditions, serviced by the Federal Medical and Biological Agency of Russia. The technique is based on hydrolysis of urine samples, extraction, concentration and derivatization with N-methyl-N-(trimethylsilyl)-trifluoroacetamide (MSTFA) followed by analysis of the sample using a combination of gas chromatography with high resolution mass spectrometry (GC-HRMS). For more confident identification and accurate determination of trace levels of 3-hydroxybenzo[a]pyrene in urine, an internal standard method and a double focusing magnetic sector mass spectrometer were used. The MM of 3-hydroxybenzo[a]pyrene was tested on a formed cohort of ten volunteers. As a result, the metrological characteristics of MM were determined and metrological certification was carried out.
