Experimental research of the biological effect of 12c and 60c ions on hepatocarcinoma cells

Brief summary

The combined effect of ionizing radiation and antitumor drugs is often used to increase the radiosensitivity of malignant cells in the treatment of resistant forms of cancer. According to WHO, one of the intractable oncological diseases is liver cancer due to the ongoing cirrhotic changes. The treatment regimen for patients with tumors of this localization includes resection, chemotherapy, targeted therapy. The current radiobiological study is based on the concept of synergy proposed by V. G. Petin and V. P. to describe the effects of the combined action of ionizing radiation and modifying agents on the survival of cell cultures. Aim. Evaluation of the effects of the combined action of 12C and 60C with a cytostatic drug on HUH-7 cells. Materials and methods. The cell lines HUH-7, HEK-293 were selected as the objects of the study. One of the impact factors was the anthracycline antibiotic doxorubicin. The cells were irradiated with 12C ions on the synchrotron U-70, as well as 60Co on the Unique Research Facility of the FSBI VNIIRAE radiation Irradiation "GUR-120". The radiation cultivation of cells consisted in carrying out a clonogenic analysis, doubling time, followed by an assessment of the synergism coefficient. Results. According to the results of experimental work, the relative biological effectiveness of the action of 12C ions with a single irradiation of normal and cancer cells was established. We have shown that a synergistic interaction is observed with the combined action of dense ionizing radiation and doxorubicin. Conclusions. Experimental data on the manifestation of the synergistic nature of the agents used on the cells of hepatocarcinoma HUH-7 were obtained.
