The effect of inhibitors of alcohol dehydrogenase on biotransformation of ethylene glycol and its esters by liver homogenate

Brief summary

The purpose of the study was to study the possibility of using homogenates of rat liver as a test system in the search for antidote therapy of poisoning by toxic alcohols. It was found that the biotransformation of ethylene glycol (EG) in the liver homogenates of rats and humans is 3-5 times faster than its methyl and ethyl esters. In turn, the rate of metabolism of EG and its esters in human liver homogenates per gram of raw tissue is faster than in rat liver homogenates. The main enzyme determining the rate of biotransformation of EG and its esters is alcohol dehydrogenase (ADH), which accounts for at least 75 and 63% of the total metabolism of these substances in human liver homogenates and non-linear rats, respectively. Ethanol and amide of isovaleric acid (AIC) inhibit the metabolism of EG and its esters in human and rat liver homogenates, and the severity of the effect depends on the concentration of both ethyl alcohol and AIC. In human liver homogenates, the suppression of biotransformation of the studied substances by ethanol and AIC was more intense than in rat liver homogenates. The similarity of the processes of biotransformation of alcohols in human liver homogenates and rats both in quality and quantity given, the latter can be used as a test systems in the search for chemicals - promising means of antidote therapy of poisoning by toxic alcohols.
